Breakthrough in spatial proteomics: Automated high-resolution LC-MS/MS tissue imaging workflow🗓
29 June, 2021⏰
8AM PDT, 11AM EDT, 4PM BST, 5PM CESTImaging mass spectrometry (IMS) approaches using MALDI IMS can produce thousands of ion images, providing molecular context to classical histological analysis. But they lack the confident linkage to protein identification and quantification. The single-cell level of sensitivity has been extended to proteome mapping using a “voxelation” approach that can enable the quantitative mapping of >2000 proteins with 100-µm spatial resolution across 12-µm thick tissue sections using a label-free quantitation approach. In addition, incorporating tandem mass tags (TMT) labeling with this workflow can improve analysis throughput, increase overall sensitivity, and enable MS imaging at 50-µm spatial resolution without compromising proteome coverage.
In this webcast, the speaker will demonstrate high-resolution and in-depth proteome imaging using an automated workflow by:✔️ Coupling a nanodroplet sample processing platform with laser capture microdissection to provide an automated sample collection system with unprecedented sensitivity
✔️ Analyzing these nanogram-quantity samples with an optimized LC system coupled to a Thermo Scientific™Orbitrap™ mass spectrometer for the sensitive, reproducible and confident analysis of the large sample sets required to create proteome maps.
✔️ The application of this innovative platform to proteome mapping demonstrates the exciting potential of proteome imaging to advance biomedical research, as described in a mouse embryo implantation study that showed a superior depth of proteome coverage and robust quantitation.
You will learn:🔻 How to optimize and automate sample preparation for low level samples
🔻 How to run large sample sets with high quality data for proteome mapping
🔻 How to quantify thousands of proteins in low level samples
🔻 How to perform bottom-up (LC-MS/MS) proteomic imaging on thin tissue sections
